Carbohydrate metabolism-Arabic

In carbohydrate metabolism we will study, the types of carbohydrate, how they are digested and absorbed, then we will the cycles of carbohydrate oxidation which generate the energy required for life…. these cycles are:

  1. Glycolysis  = Glucose oxidation (i.e. break down of glucose to get energy).

  2. Frucose metabolism.

  3. Galactose metabolism.

  4. Gluconeogensis = formation of Glucose form non-carbohydrate sources.

  5. Kerbs cycle.

  6. Glycogen Metabolism including breaking and formation of glycogen.

  7. Pentose phosphate pathway = which generate NADH

فالميتابولزم بتاعة الكربوهيدرات احنا هنعرف انواع الكربوهيدرات وطريقة هضمها وامتصاصها وبعد كدة هنعرف السيكلز بتاعة اكسدة الكربوهيدرات للحصول عالطاقة وهم:

1-     Glycolysis وهى اكسدة الجلوكوز

2-     Fructose metabolism

3-     Galactose metabolism

4-     Gluconeogensis وهى تخليق الجلوكوز من مركبات مش سكرية

5-     Kerbs cycle

6-     Glycogen metabolism وتشمل تكسير وتكوين الجليكوجين

7-     Pentose phosphate pathway ودى السيكل البتعمل NADPH وهنعرف ليه وفايدتها ايه قدام

وبعد كدة هنتكلم عن الdiabetes mellitus.

A) Types of carbohydrates:

1. Monosaccharides

-   They are those carbohydrates that cannot be hydrolyzed into simpler carbohydrates.

-   They may be classified as trioses (3-carbon sugar), tetroses (4-carbon sugar), pentoses (5-carbon sugar), hexoses (6-carbon sugar), or heptoses (7-carbon sugar).

2. Disaccharides:are condensation products of two monosaccharide units. Examples are maltose and sucrose.
3. Oligosaccharidesare condensation products of 2 to 10 monosaccharides; Example: maltotriose
4. Polysaccharidesare condensation products of more than ten monosaccharide units; examples are the starches and dextrins, which may be linear or branched polymers.

 دى سهلة مشمحتاجة شرح ولا توضيح ولو مشواضحة اكتب كومنت فاخر الموضوع وهنوضحها

B)   Carbohydrate digestion:

After eating, digestion begins as follow:

1) In the mouth, salivary amylase, hydrolyze starch partially into a mixture of dextrins and maltose.

2) In the stomach, salivary amylase continues hydrolysis of starch only for few minutes then stop because the pH becomes acidic due to the presence of the HCl in stomach and this is unfavourable condition for the amylase to work.

3) In the intestine, pancreatic amylase, completes the digestion of starch into maltose with little isomaltose and maltotriose which are then hydrolyzed in the intestine into glucose. Fructose and Galactose.

بعد الاكل يتم الهضم كالاتى:

1) فى الفم، انزيم الامايليز (Salviary amylase) يتم تكسير النشا جزئيا الى dextrins وMaltose.

 2) فى المعدة، الSalivary amylase بيكمل شغله يعنى بيستمر فى تكسير النشا ولكن لدقايق قليلة وبعد كدة يتوقف عشان الوسط الحامضى اللى فالمعدة مشمناسب ليه.

3) فى الامعاء، الامايليز بتاع البنكرياس (Pancreatic amylase) بيكمل تكسير النشا الى maltose، وIsomaltoseوMaltotriose واللى فالنهاية بيتكسروا فالامعاء الى Glucose وFructose وGalactose. ذى ما هنشوف فالمعادلات اللى جاية

Starch + H2O   ===== Amylase ====>  Dixterns + Maltose

Dixterns + H2O  ===== Amylase ====> Maltose + Isomaltose + Maltotriose

Maltotriose + H2O ===== Maltase====>Maltose + Glucose

 Maltose + H2O  ======== Maltase====> Glucose + Glucose

Isomaltose + H2O===== Isomaltase====> Glucose + Glucose

Sucrose + H2O========== Sucrase=====>Glucose + Fructose 

Lactose + H2O ========Lactose=======>Glucose + Galactose

4. Sucrose and the enzymes that complete hydrolysis into Monosaccharides in the mucus layer of the intestine

5. The net result of carbohydrate hydrolyses is glucose, fructose and Galactose

4) السكاروز (sucrose) والانزيمات اللى بتكمل اكسدة السكريات الاحادية اللى اسمها Monosaccharides بتحصل فى عضلات الامعاء

 5) نهاية تكسير الكاربوهيدرات بتكون glucose, fructose and Galactose

C) Absorption:

دلوقتى احنا عرفنا من فوق اذاى هضم الكربوهيدرات بيحصل ودلوقتى هنعرف الامتصاص ودخول الجلوكوز فى  الخلايا بيحصل اذاى.

- The polysaccharides and oligosaccharides are not absorbable so, they must be converted to Monosaccharides.

- Monosaccharides are principally absorbed from the duodenum then pass into the blood through the hepatic portal vein to the liver where:

  • Part of these monosaccharides  is stored asglycogen and part is oxidised through glycolysis to obtain energy

  • Part is oxidised through the pentose phosphate pathway to regenerate NADPH which, together with glucose itself, is used in synthesis of such molecules as amino acids, nucleotides, fats and cholesterol

  • Part is oxidised to produce energy (ATP) which is used in the anabolism processes.

- بعد ما هضم الكاربوهيدات المعقدة الى سكريات احادية واللى هما الجلوكوز والفركتوز والجالاكتوز، بيتم امتصاص السكريات الاحادية فى الاثنى عشر وبعد كدة بيمشى فالدم ويوصول للكبد عن طريق الوريد الكبدى البابى وفالكبد بيحصله الاتى:

       ===> جزء من السكريات الاحادية بيتخزن فى صورة جليكوجين وجزء بيتأكسد فى الGlycolsis عشان ناخد طاقة منه.

      ===> جزء تانى بيتاكسد بس عن طريق الpentose phosphate pathway عشان ناخد منه NADPH اللى بيستخدم مع الجلوكوز لتكوين جزيئات معينة من الاحماض الامينية ونيوكليوتايد اللى بتستخدم فتخليق الDNA ودهون وكوليسترول

===> جزء تانى بيتاكسد عشان نعمل ATP وده طريق لتخزين الطاقة والطاقة المخزونة دى بنستخدمها فى بناء خلايا جديدة او تكوين اى جزيئات او مواد ضرورية فالجسم

Before we know the mechanism by which the glucose is absorbed into the cells we must know the composition of the cell membrane through which the glucose pass.

this part will help any any one who don’t study biology or cell before to be able to understand biochemistry

قبل ما نعرف الطريقة اللى بيدخل بيها الجلوكوز فى الخلايا لازم نعرف الاول تركيب غشاء الخلية وده جزء مهم للناس اللى مدرستش بيولوجى ولا تركيب الخلايا قبل كدة يعنى ده عشان

Composition of the cell membrane:

==>  As from the previous picture, the cell membrane is composed of 2 layers each of them consists of a layer of phospholipids.

==>  Each phospholipid molecule consists of apolar hydrophilic head and a non-polar called hydrophobic tail.

من الشكل السابق، شايفين ان غشاء الخلية بيتكون من طبقتين كل واحدة فيها بتتكون من طبقة من الphospholipids وكل جزئ من الphospholipids بيتكون من راس او جزء محب للماء (hydrophilic) وطبعا كلمة محب للماء دى معناها انها مشحونة وبعد كدة بيتكون من سلسلة كربونية طبعا غير مشحونة لان الكاربون بيكون غير مشحون وبنسمى السلسة دى الزيل الغيرمحب للماء (hydrophobic tail)  …. وده موضح فالصورة اللى جنب الكلام

==> How the bilayer membrane is formed: And as we knowthe extracellular and intercellular fluid are polar, therefore the polar heads are arranged towards the polar fluids and the non-polar tails arranged toward themselves  inward the membrane where there are a hydrophobic interaction between them and thus forming a phospholipids bilayer membrane

==> And across the membrane there are transport proteins and receptor proteins

هنا بقى بيقول ايه …. اذاى بيتكون غشاء الخلية من طبقتين يعنى ممكن نقولها بطريقة تانية … اذاى الطبقتين دول ماسكين فبعض بالطريقة دى من غير ماينفصلو عن بعض:- طبعا ذى ما احنا عارفين ان السائل اللى خارج الخلية واللى داخل الخلية هما ال2 polar يعنى مشحونين وده يعمل ايه …. يخلى الراس المشحونة او الجزء المشحون فى جزيئات الphospholidis هى اللى بتحب تعوم فالسائل المشحون وطبعا الزيل اللى مشمشحون مشهيقدر يعوم فالسائل المشحون فيعمل ايه .. الديول اللى مشمشحونة يحضونو بعض ويخبو بعض من السائل المشحون طيب هيخبو بعض اذاى؟ …. هيعمل الطبقتين بالطريقة اللى شايفانها فالصورة يعنى دى طريقة لانهم يهربو من السائل المشحون بس كدة …. وفيه كمان بروتينات ناقلة ومستقبلات بروتينية (receptors) ودى هنعرف وظيفتها فالحاجات اللى جاية

==> Function of the cell membrane:-

  • Protection of cells.

  • On the other hand, it represents a barrier which prevents entry of some molecules into cells such as polar molecules but it uses other mechanism by which the molecules enter the cells such as Na-K pump through which ions are transported into the cell.

    ايه هى وظيقة غشاء الخلية ده:

    - حماية الخلية من الوسط المحيط بيها

    - وعلى ناحية تانية … هو عبارة عن حائل او حاجز بيمنع دخول بعض الجزيئات فى الخلية ذى الجزيئات الpolar اللى عليها شحنة ولكن بيستخدم طرق تانية عشان يدخل الجزيئات دى بكميات معينة على حسب ماتحتاج الخلية ذى مثلا ممكن يدخلها عن طريق حاجة اسمها Na-K pump اللى عن طريقها بيدخل الايونات فى الخلية

In case of glucose:

Any polar molecules can’t pass through the inner non-polar layer of cell membrane so, how glucose enter the cell through the cell membrane while it is polar?

===> The answer is that glucose has 2 mechanism to enter the cells:

بالنسبة للجلوكوز:

طبعا احنا عارفين ان الجزيئات المشحونة متتفقش ابدا مع الجزيئات الغير مشحونة …. طيب الغشاء بتاع الخلية من جوة متكون من الديول الغير مشحونة يبقى اذاى الجلوكوز المشحون هيعدى منهم … الاجابة ان فى طرقتين تعالو نشوفهم.

1) The passive diffusion or transport:

===> DEF: it transport of biochemical and other atomic or molecular substance across the cell membrane into the cell from higher concentration region to a lower concentration region without any need for energy. (i.e. the substance enter the cell only if its concentration outside the cell is more than its concentration inside the cell.)

===> It depends on the concentration gradient.

===> It doesn’t need energy.

===> Depends on the permeability of the cell membrane.

===> Types:

  • Simple diffusion:in which the substance are transported from higher concentration to lower concentration region through the phospholipids bilayer without any need for energy and without using the transmembrane proteins (carriers/transporters/channels/pores).

  • Facilitated diffusion or passive-mediated-transport:in which the substance are transported from higher concentration to lower concentration region through the phospholipids bilayer using the transport proteins  and without any need for energy.

-          Fructose and pentose use this mechanism.

-          Glucose is transported by this mechanism into brain, kidney and liver.

1. الPassive diffusion or transport:

لو ترجمنا كلمة passive على اى قاموس هنلاقيها بعنى بليد او سلبى او غير فعال لكن هنا مشهنترجمها كدة لكن هنسميها السهل يعنى العنوان هيكون ترجمته النقل السهل …. طيب سهل اذاى …. عشان هو عبارة عن نقل الذرات او الجزيئات خلال غشاء الخلية الى داخل الخلية لما بيكون تركيز المواد دى خارج الخلية اعلى من داخل الخلية عشان كدة سموه سهل يعنى ذى ما نهر النيل بيتحرك من المكان العالى للمنخفض وطبعا ده مشمحتاج طاقة وبيعتمد اساساى على تركيز المواد لانها لازم تكون خارج الخلية اعلى من داخل الخلية

وليه نوعين

 ====> الاول ….. النوع البسيط (simple diffusion) …. وده مجرد انتقال المادة من تركيز عالى خارج الخلية الى تركير منخفض داخل الخلية خلال الphospholipid bilayer بدون استهلاك طاقة او حتى استخدام بروتينات ناقلة

====>التانى ….. النوع النقل اللى بيتم تسهليه (facilitated diffusion) …. وده مجرد انتقال من تركيز عالى خارج الخلية الى تركيرز منخفض داخل الخلية خلال الغشاء عادى وبدون استهلاك طاقة لكن بمساعدة بروتينات ناقلة يعنى فى وسيلة مساعدة او تسهيل لعملية النقل

الpassive transport ده مين بيستخدمه … الفركتوز والبنتوز (السكر الخماسى) والجلوكوز بيستخدمه برضو بس فى حالة دخوله خلايا المخ والكلية والكبد

2) Active diffusion or transport

===> DEF: it means transport of a substance from region of lower concentration to a region of higher concentration (i.e. against the concentration gradient) utilizing energy from ATP molecules and carrier proteins.

===> It doesn’t depend on the concentration gradient.

===> It needs energy.

===> This mechanism applied by the cell to accumulate high concentration of molecules that the cell needs such as ions, glucose and amino acids.

===>Doesn’t depend on the permeability of the cell membrane because this mechanism transports the substance through the transport proteins and carrier proteins.

===> It has 2 types:

  • Primary active transport: in which the cell uses chemical energy such as ATP.

  • Secondary active transport: in which the uses electrochemical gradient such as sodium and potassium dependent ATP bump

===> It is the mechanism by which glucose is transported from intestinal tract

النوع التانى الactive diffusion or transport:

كلمة active احنا عارفين ان معناها نشيط … طيب سموه كدة ليه …. عشان ده عبارة عن نقل المادة من تركيز منفخض خارج الخلية الى تركيز عالى داخل الخلية يعنى بينقل المادة فى عكس تدرج التركيز وده طبعا محتاج طاقة لانه عامل ذى واحد بيزق حاجة وعايز يدخلها فمكان مليان قوى واكيد هيزقها بالعافية يعنى متحاج طاقة وبياخد الطاقة دى من جزيئات الATP وطبعا الطريقة دى لا تعتمد على تركيز المادة لانه مشفارق معاه المادة تركيزها ايه لانه بينقلها بالطاقة

وفى حاجتين ممكن نستنتجهم لوحدنا وهما ايه …. الاولى …. ان الطريقة دى بتستخدمها الخلية لعمل تراكم من جزيئات معينة داخل الخلية عشان هى محتاجها بكيمات كبيرة وده ذى الجلوكوز لان الخلية علطول بتحرق الجلوكوز عشان تاخد طاقة …… والتانية ….. ان الطريقة دى لا تعتمد على مدى نفاذية الغشاء للمادة لانها بتنتقل خلال البروتينات الناقلة

وفى نوعين من الطريقة دى:

===> النوع الاولي …… وده فيه الخلية الطاقة اللى بتستخدمها فعمليه النقل بتاخدها من جزيئات الATP

===> النوع الثانوى …… وده بتسخدم الطاقة بتاعة الATP مع الNA-K Pump

Glucose transportation into the cell by insulin action:

===> When the blood glucose level is high, the nervous system sends signals to the beta-cells of the pancreas to secrete insulin.

===> Then insulin binds to its receptors on the outside surface of the cell membrane causing conformational changes to the receptors (where the receptors are soft protein) leading to conformational changes to the cell membrane and opening of protein gates which called glucose transporter (GLUT) leading to entrance of glucose inside the cell.

===> Opening of these gates lead to activation or deactivation of some enzymes that responsible to glucose oxidation.

===> Insulin binding to the receptor is a reversible process because it will leave the receptor after delivering the message.

===> This process is regulated by the central nervous system.

===> Then after the glucose level returns normal; the insulin leave the receptor and no glucose will enter the cell

===> But in some cases the B-cells is highly activated secreting more insulin which make the glucose to enter the cell and  its level is lowered causing hypoglycaemia

===>The receptors of the insulin are specific for insulin and are distributed in all tissue with different concentration where the receptor concentration increases in the tissue:

  • Which utilize glucose as the main source of energy such as brain and cells of the nervous system, red blood cells, muscles,…..etc

  • At which the blood supply is low such as the peripheral tissues.

لما يعلى السكر فى الدم، الجهاز العصبى يرسل رسائل الى الB-cell فى البنكرياس عشان يفرز الانسولين

والانسولين بيروح على الخلية ويمسك ف الreceptor بتاعه وده يعمل تغير فشكل الreceptor عشان هو بروتين طرى يعنى soft وده بيعمل تغير فشكل الغشاء والتغير ده بيفتخ بوابات  بروتينية (protein gates) معينة على سطح الخلية بنسميها ناقلات الجلوكوز (glucose transporter) ودى بتسهل دخول الجلوكوز وكمان فتح البوابات دى بيعمل تنشيط او تثبيط لبعض الانزيمات البتعمل اكسدة للجلوكوز وبالتالى تبدا الglycolysis اللى هى أكسدة الجلوكوز وبعد كدة الانسولسن يسيب الreceptor لما مستوى السكر يقل ومفيش جلوكوز هيدخل تانى لكن فى بعض الحالات بتكون الB-cell نشطة وتفرز انسولين بتركيز عالى وبالتالى الجلوكوز هيدخل فالدم بالرغم انه قليل ويعمل Hypoglycaemia اللى هى نقص فى مستوى السكر فالدم.

تركيز الreceptor بيختلف على حسب احتياج الخلية للجلوكوز وكمان على حسب وصول الدم بمعنى ان الاعضاء المحتاجة جلوكوز كغذاء اساسى ذى المخ والاعصاب بيزيد تركيز الreceptor فيها والعكس صحيح طبعا وكمان الاماكن التى يقل فيها وصول الدم برضو ذى الاطراف يزيد فيها تركيز الreceptors دى

Why glucose is stored in the liver and muscles as glycogen and not as it is (i.e. glucose)?

Because glycogen is solid polymer that is compacted and occupy a small size inside the cell which prevents cell membrane rupture by the pressure on the cell membrane.

While if the glucose is stored as glucose, it will take large size which then cause pressure on the cell membrane and then rupture of the cell membrane.

بالعربى الجليكوجين بيكون مضغوط وواخد حجم صغير فالخلية لكن الجلوكوز لما بيتراكم فوق بعضه حجمه بيكون كبير وممكن يضغط على غشاء الخيلو ويقطعه.

What is the factor that determines the amount of glucose that enters the cell?

The amount of glucose after meal where the glucose must be maintained at the normal level and the excess must enter the cells

بالعربى انه لما السكر بيزيد فى الدم بعد الاكل الكمية الزيادة هى البتدخل فى الخلايا يعنى البيحدد كمية الكلوجوز الداخلة فى الخلايا هو كمية الجلوكوز الداخل مع الاكل بحيث ان الزيادة فى الدم عن التركيز الطبيعى هو اللى بيدخل فالخلايا حتى لو عندها سكر كتير وده طبعا بيستخدم الactive transport.

Is the metabolism of fats can be used in formation of glycogen?

The answer is yes because the result of oxidation of fat is formation of glycerol which can enter the gluconeogensis process in which glucose is formed.

So, if glucose can be formed from fats thus glycogen can be form fats

هنا بيسال هل تكسير الدهون ممكن نون منه جليكوجين …. الاجابة ايوة ممكن … عشان من نواتج اكسدة الدهون هو الجليسرول وده بيدخل فى الدورة بتاعة الgluconeogensis اللى هى تكوين الجلوكوز من مواد غير سكريه او كاربوهيدراتيه وطالما نقدر نكون جلوكوز من الدهون يبقى اكيد نقدر نكون جليكوجين من الدهون

D)  Structure of glucose:

===> Glucose is aldose sugar and has 2 isomers: (D-glucose and L-Glucose)

It has 2 chiral carbon atoms (i.e. the carbon atom that carry 4 different groups); the carbons number 2 and 3

هنا عايز يعرفنا ان الجلوكوز ليه شكلين فالفراغ …. الشكل L والشكل D وطبعا ده عشان جزء الجلوكوز فيه كاربونة كايرال يعنى ايه….. يعنى كاربونة عليها 4 مجموعات مختلفة

The (L)-glucose is the isomer that is utilized by the cells. WHY?

Because the amino acid molecules which forms the enzyme present in the L- form which make the enzymes in the L- form, therefore the L-glucose is the more suitable substrate which is the more matching with the binding groups in the active site than the D-glucose

Proteins molecules also have L- and D- forms because all has a chiral carbon except glycine which doesn’t carry 4 different groups.

بالعربى كدة ان البروتينات اللى بتكون الانزيمات موجودة فى ال L-form وبالتالى الانزيمات الموجودة هتكون فال L-form وبالتالى الL-glucose هيكون هو الsubstrate المناسب

How the scientists classified the isomers into L- and D- forms?

They use the glyceraldehyde as a standard as follow:

-          Each isomer has a chiral carbon (i.e. the carbons atom which carries 4 different groups)

-          The isomer that has L- and D- forms are called enantiomers

هنا بيعرفنا اذاى العلماء قسمو المركبات من حيث شكلها الفراغى الى L-form والى D-form وبيقول انهم استخدمو الجليسرالدهيد كمركب قياسى يقيسو عليه باقى المركبات كالاتى:

====> كل شكل فراغى ليه كاربونة Chiral يعنى تحمل 4 مجموعات مختلة

====> والL-Form والD-form هما ال2 بنسميهم enantiomers اللى ترجمتها بالعربى المتبلورات المضادة يعنى كل واحد فيهم عكس التانى ذى كف ايدك لما تحطهم قدام عينك هما ال2 شبه بعض بس معاكسين لبعض بمعنى انك لما تحطهم قصاد بعض هتلاقى الابهام فايدك اليمين ناحية اليمين واللى فايدك الشمال  ناحية الشمال لكن مستحيل تلاقى ال2 فاتجاه اليمين او ال2 فاتجاه الشمال

Differentiate between the following terms:

1.      Enantiomers:

Are 2 stereoisomers that are mirror images of each other that are “non-superposable” (not identical), much as one’s left and right hands are “the same” but opposite

الenantionmers دى مركبات متشابها فراغيا ولكن كل واحد منهم كانه صورة فالمراية من التانى لن لا يطبقو على بعض ذى ماقولنا فوق على كف الايد مينفعش تلاقى صوابع الابهام فايديك ال2 فاتجاهين مختلفين لكن مستحيل تلاقيهم فاتجاه واحد

2.     Epimers:

Epimers are diastereomers that differ in configuration of only one carbon atom and they are non-superposable, and non-mirror images of one another

The glucose molecules are non-mirror image to each other but aren’t identical because they differ in one carbon atom.

الepimer ده برضو متشابهات فراغية ولكن تختلف فى كاربونة واحدة وهما ال2 لا ينطبقو على بعض ومحدش فيهم كانه صورة فالمراية من التانى

3.    Anomers:

In carbohydrate chemistry, an anomer is a special type of epimer because it is a stereoisomer of a cyclic saccharide that differs only in its configuration at the hemiacetal or hemiketal carbon, also called the anomeric carbon

The cyclic structure of glucose has 2 anomers because the hydroxyl groups orientation differes on the hemiketal carbon atom.

الAnomers دى حالة خاصة من الepimer بمعنى انها مركبات متشابهة فراغيا وتختلف فكاربونة واحدة ومحدش فيهم صورة فالمراية للتانى ولكن بتحصل فالسكريات الخلقية وبيختلفو فالكاربونة شبيه الالدهيد او شبيهة الكيتون اللى بنسميهم بالانجليزى hemiacetal و hemiketal

والجلوكوز ليه anomer 2 عشان وضع الهيدروكسيل OH  بتاعة الى الhemiketal carbon بتختلف فالفراغ ذى ماشيفين فالصورة



احنا طبعا شوفنا فى المحاضرة الالولى اذاى حصل هضم للكربوهيدرات وان فى النهاية بيكون عندنا glucose, fructose andGalactose وتتبعنا مسار الجلوكوز وعرفنا انه بيروح للكبد عن طريق الوريد الكبدى البابى والمفروض هيحصله اكسدة فالكبد والعملية دى بنسميها glycolysis يعنى تحلل اوتكسير الجلوكوز وطبعا ده عشان نحصل على طاقة

قبل ما نبدا لازم نعرف الناس ايه هو الATP ده اختصار الى Adenosine Tri-Phosphate  وده ببساطة هو عملة الطاقة فالخلية والجسم كله ….اذاى؟ …. عشان فيه 3 جيزئات فوسفات كل واحدة فيهم لما تتكسر بدى طاقة كبيرة قوى ولذلك لما اجى اقول ان الخطوة رقم واحد مثلا اخدت جزئ فوسفات من جزئ ATP ده معناه ان الخطوة دى استهلكت طاقة

Glycolysis

A)  Definition:

It is the breakdown of glucose in the cell cytosol (= cytoplasm) producing pyrurate in the presence of oxygen or lactate in the absence of oxygen.

 هى تكسير الجلوكوز فى السيتوبلازم عشان يدينا pyruvate فى وجود الاكسجين او lactate فى غياب الاكسجين

B)   Site:

Glycolysis occurs in cytoplasm but:

  • In presence of O2, oxidation of glucose is complete in mitochondria where pyruvate enters the kerb’s cycle and the electron transport chain to complete the oxidation of glucose resulting in a high amount of energy.

  • In absence of O­2, pyruvate is converted into lactate in the cytoplasm giving a small amount of energy but it is important to some tissue.

Therefore:

Occurrence of glycolysis is of physiological importance in:

  1. Tissues with no mitochondria such as RBCs, cornea and lens.

  2. Tissues with few mitochondria: Testis, leucocytes, medulla of the kidney, retina, skin and gastrointestinal tract

  3. Tissues undergo frequent oxygen lack: skeletal muscles especially during exercise.

Where they depend only on the glycolysis not on kerbs cycle and the electron transport chain

الglycolysis اللى هى اكسدة الجلوكوز بتحصل فالسيتوبلازم ولكن ممكن تحصل فى وجود الاكسجين او فى غيابه بس فى حالة انها تحصل فى وجود الاكسجين لازم الpyruvate يروح الميتوكوندريا عشان يدخل فى الkerbs cycle والelectron transport chain عشان نكمل اكسدة الجلوكوز ونحصل منه على طاقة عالية … طيب اذاى يعنى … هو مشالglycolysis هى اكسدة الجلوكوز طيب اذاى هنكملها فسيكل تانية؟ …. الpyruvate ده عبارة عن 3 كاربون من جزء الجلوكوز والglycolysis بتنتهى عند تكوين الPyruvate بمعنى ان فى الglycolysis بيحصل اكسدة ل3 كاربونات فقط وبنحصل على طاقة من3 كاربونات فقط ولذلك الpyruvate اللى هو ال3 كاربونات الباقية بتكمل اكسدته فى الميتوكوندريا

ولكن فى حالة غياب  الاكسجين يتحول الpyruvate الى lactic acid or lactate ويدينا كمية طاقة قليلة جدا بس برضو مهمة لبعض الانسجة  ذى مثلا:

===> الانسجة اللى مفهاش ميتوكونديريا بتعتمد على كمية الطاقة القليلة دى عشان مشقدامها غير انها تحول الpyruvate الى الlactate   والانسجة اللى مفهاش ميتوكوندريا ذى كرات الدم الحمراء والقرنية والعدسات

===> ذيهم برضو الانسجة اللى فيهم عدد قليل من الميتوكوندريا ذى الطبقة اللى فيها النيفرونات فالكلية وكرات الدم البيضا والخصيتين وشبكية العين والجلد والجهاز الهضمى

===> كمان الانسجة اللى بتتعرض الى نقص اكسجين بصورة متكررة ذى مثلا العضلات وقت التمارين العنيفة …. طيب اذاى؟ …. مثلا فحالة التمارين الكتير العضلات تحتاج الى كمية طاقة كبيرة جدا وبالتالى الامداد بالاكسجين بيكون غير كافى فيكون قليل بالنسبة لكمية الاحتياج فلما يتاكسد الجلوكوز ونحصل منه على pyruvate هيتحول الى lactic acid عشان الاكسجين قليل وبالتالى الacidity هتزيد والpH هتقل وده هيصيب العضلة بالاجهاد وبالتالى فنقص او غياب الاكسجين الخلية تعتمد على الglycolysis فالسيتوبلازم ولاتعتمد على kerbs cycle فالميتوكونديريا.

C)    Stages of glycolysis:

الglycolysis تتم فى 10 خطوات على مرحلتين كل مرحلة 5 خطوات واول ورحلة بتستهلك طاقة والتانية بتدى طاقة.

وطبعا الglycolysis تتم بعد فتح الgates  القولنا عليها عن طريق الانسولين

It is the energy requiring stage

in this stage:

  • One molecule of glucose is converted into two molecules of glyceraldehyde-3-phosphate.

  • These steps consume 2 molecules of ATP .

Step one:

1.      Event: (glucose phosphorylation)­

===> A phosphate group is transferred from ATP molecule to the carbon number 6 in the glucose molecule forming glucose-6-phosphate, thus this step is energy consuming.

===> Glucose-6-phosphate is an intermediate forming an important branch point in the metabolism.

===> This step is fast irreversible step

يعنى جزء الجلوكوز بياخد مجموعة فوسفات فى الكربونة رقم 6 من جزء ATP البيتحول الى ADP وده معناه ان الخطوة دى بتستهلك طاقة والخطوة دى فتكون فاتجاه واحد يمعنى ان النواتج مينفعش يتفاعلو مع بعض تانى ويدونى جلوكوز

2.  Enzymes stimulating this step:

===> The enzymes used are either glucokinase or hexokinase enzymes which are responsible for entry of glucose into the cell and phosphorylation of glucose which lead to glucose trapping inside the cell, therefore this step is irreversible because if it is reversible the glucose-6-ph will return to glucose and could exit from the cell again.

===>They are activated after a carbohydrate rich meal for 2 hours to lower the glucose blood level.

NOTE: Kinase enzyme always add phosphate group on the substrate

بمعنى الانسولين يفتح الgates ذى ذى ماقولنا فى المحاضرة الاولى وينشط الانزيمات دى هتنشط وتساعد الجلوكوز عالدخول وتعمله phosphorylation اللى بتعمل trapping للجلوكوز يعنى بتحبسه جوة الخلية …. اذاى بقى؟؟ …. مجموعة الفوسفات هتذود الpolarity بتاعة الجلوكوز يعنى هتذود الشحنة بتاعته فهتخليه ميعرفش يخرج عشان مشهيعرف يعدى فى الجزء الnon-polar بتاع غشاء الخلية ذى ماقولنا المحاضرة الاولى برضو وبالتالى ميعرفش يخرج تانى من الخلية والgates مفتوحة وعشان كدة الخطوة دى سريعة و irreversible يعنى فاتجاه واحد عشان لو كانت فاتجاهين الg-6-p هيتحول لجلوكوز تانى وبالتالى الجلوكوز ممكن يخرج من الخلية والgates مفتوحة لان الgates دى هتفضل مفتوحة شوية لحد ماتدخل كمية من الجلوكوز يعنى هى مشبتّدخل  جزئ واحد وتقفل وراه وبالتالى لو معملناش الPhosphorylation وحبسنا الجلوكوز هيعرف يخرج تانى

ٍStep 2:

1.   Event: (formation of fructose-6-phosphate from glucose-6-phosphate)­

Isomerization of glucose-6-phosphate to fructose 6-phosphate, I.e. a conversion of an aldose into a ketose by phosphoglucose isomerase.

It doesn’t need energy because it occurs spontaneosly.

2.   Enzymes stimulating this step: phosphoglucose isomerase.

NOTE: Isomerase enzyme always catalyzes the structural rearrangements.

فى الخطوة دى انزيم الisomerase الى وظيفته يعمل تعديل  فى شكل الجزء…. بيعمل ايه؟ …. بيشيل جزئ هيدروجين من الكربونة رقم 2   يعنى ذرة من الكربونة وذرة من الOH ويحول ال(C-OH) الى (C=O) ويحط جزئ الهيدروجين على الكربونة رقم 1 اللى هى مجموعة الالدهيد (CHO) يعنى هيحط ذرة هيدروجين على الكاربونة وواحدة على الO ويحول ال (H-C=O) الى(CH2OH)

Step 3:

a.  Event: (Phosphorylation of Fructose-6-phosphate to fructose-1,6-­bisphosphate)­

-  Phosphorylation of Fructose-6-phosphate by ATP to fructose-1,6-­bisphosphate (F-1 ,6-BP)

-   This step is irreversible.

b.  Enzymes stimulating this step: by phosphofructokinase (PFK)

طبعا قولنا فالخطوة الاولى ان الkinase بيضف مجموعة فوسفات على الجزئ وهيجيب الفوسفات ده منين؟ …. اكيد من جزئ ATP يعنى هيستهلك طاقة على الجزئ واسم الانزيم phosphofructokinase عشان هيضيف مجموعة فوسفات على الفركتوز بس كدة

step 4:

a. Event: (cleave of 6-carbon sugar into two 3-carbon fragments)­

-  Splitting of fructose-1,6-bisphosphate into tow 3-carbon fragments:

  • Glyceraldehyde 3-phosphate (GAP)

  • Dihydroxyacetone phosphate (DHAP)

-   Reversible under intracellular conditions

b.  Enzymes stimulating this step:

-  Aldolase. (This enzyme derives its name from the nature of the reverse reaction, an aldol condensation).

طبعا دى سهلة انه مجرد بيكسر الجزء الى مركبين كل واحد فيهم 3 كربونات والانزيم ده اسمه aldolase متاخد من اسم التفاعل العكسى اللى هو ال aldol condensation اللى فيه بيفاعل كيتون مع الدهيد فسموه aldolase

Step 5:

a.  Event: (isomerisation of (GAP) to (DHAP))­

-  Isomerisation of Glyceraldehyde 3-phosphate (GAP) to Dihydroxyacetone phosphate (DHAP)

-  It is fast reversible step.

b. Enzymes stimulating this step: triose phosphate isomerase (TPI or TIM).

c.  Importance of this step:

To get full energy from glucose molecules in other words to get energy from the 6 carbons.EXPLAIN?

-   The cell can’t oxidize DHAP to get energy from it , thus it will be lost as a waste product.

-    Thus, the cell will obtain energy from GAP only (i.e. from 3 carbon of the glucose), not from the 6-carbons.

-    But, the GAP will continue the glycolysis and give energy, while DAHP is not

-     Thus, in order to get energy from the 6-carbons of glucose, the DHAP must be converted to GAP

الDHAP ده مشمفيد وعبارة عن waste بالنسبة للخلية ولو الخلية اتخلصت منه يبقى هتاخد طاقة من 3 كربونات بس مش من الستة كلهم وبالتالى الافيد ان الDHAP يتحول الى الGAP عشان الGAP بيكمل الglycolysis

After this step we get 2 molecules of Glyceraldehyde 3-phosphate (GAP):

One form step 4 and one form step 5.

طبعا كدة هيكون عندنا جزيئين GAP واحد من الخطوة الرابعة وواحد من الخطوة الخامسة وخلينا فاكرين ده عشان لما نيجى نحسب جزيائات الATP فى الاخر نعرف نحسبهم صح

(The energy producing stage)

In this stage:

Tow moleculse of Glyceraldehyde 3-phosphate (GAP) is converted into two molecules of pyruvate producing 4 molecules of ATP.

Step 6

a.  Event: (Oxidation of glyceraldehyde 3-phosphate to 1,3-bisphosphoglycerate)

Conversion of glyceraldehyde-3-phosphate into 1,3-­bisphosphoglycerate (1,3-BPG), a reaction catalyzed by glyceraldehyde-3-phosphate dehydrogenase.

b.  Enzymes stimulating this step: glyceraldehyde 3-phosphate dehydrogenase.

Note that, the phosphate used in this step is inorganic phosphate.

The oxidation of the aldehyde to an acid is coupled to the reduction of NAD+ to NADH/H+

الNAD هنا فايدته عامل مؤكسد لان ذى ماكلنا عارفين ان كل عملية اكسدة لازم يقابلها اختزال وبالتالى الNAD هيحصلها اختزال بانها تاخد جزئ الهيدروجين من مجموعة الCHO بتاعة ال Glyceraldehyde 3-phosphate (GAP)


Importance of 1,3-bisphosphoglycerate:

- During the glycolysis inside the RBCs, part of the 1,3-bisphosphoglycerate is converted into 2,3-bisphosphoglycerate by the enzymebisphosphoglycerate mutase

-   2,3-BPG decreases the affinity of Haemoglobin for Oxygen (i.e. decreases the attachment of O2to haemoglobin) thus the oxygen can leave hemoglobin easily and this is good in case of oxygen shortage because cells will be able to get their needs of O2 easily even if the oxygen supply is low.

اثناء الglycolysis جزء من ال (1,3BPG) بيتحول الى (2,3BPG) …. طيب وايه يعنى هيفدنى بايه؟ ….. ال2,3BPG بيقلل قابلية الهيموجلبين فى كرات الدم الحمرا على الارتباط بالاكسجين وده معناه ان الاكسجين يقدر يسيب الهيوجلوبين بسهولة جدا وده هيفيد امتى؟… لما نكون فى منطقة مفهاش اكسجين عشان الخلايا بتبقى عايزة اكسجين كتير وتعالو نتخيل لو ال2,3BPG مشموجود والاكسجين قليل هيحصل ايه؟….. ارتباط الاكسجين بالهيموجلوبين هيكون قوى وده معناه ان الخلايا هيبقى صعب انها تاخد الاكسجين يعنى يبقى الاكسجين قليل والخلايا مشقادرة تاخده وال2,3BPG هو اللى بيحل المشكلة دى عن طريق انه يسهل على الخلايا انها تاخد الاكسجين.

Clinical and physiological aspects of 2,3-BPG تطبيقات للكلام اللى قولناه

  1. smokers and people who live in high altitude, where the Hb increase in each RBC and the number of RBCs increase and increase the amount of 2,3-bisphosphoglycerate leading to decrease the affinity of Hb for O2 causing dissociation of O2 form Hb easily into the blood capillary and thus the cells takes its need easily.

  2. Fetus gets oxygen form the mother thus fetal Hb has a high affinity for O2 because it has a plenty of O2 from his mother, thus competing with the Hb of the mother because the oxygen leave the Mother Hb because of it low affinity and attach to the Fetus Hb which has a high affinity for the O2.

  3. The sotred blood for blood transfusion has diminished levels of 2,3-BPG, thus before transfusion of inosine or the glycolytic substrate dihydroxyphosphate to regenerate 2,3-BPG before blood transfusion because in absence of 2,3-BPG suffocation اختناق occurs.

ذى الناس المدخنين او اللى فى المناطق العالية الاكسجين بيبقى قليل فالمادة دى بتذيد عندهم عشان يسهل انفصال الاكسجين من الهيموجلوبين وبالتالى الانسجة تحصل عالاكسجين بسهولة

والجنين الاكسجين بيجيله من الام بسهولة لان الهيموجلوبين بتاع الام ارتباطه بالاكسجين قليل ويروح للجنين يعنى الجنين بيجيله الدم بسهولة وكتير يعنى معندوش نقص اكسجين فمشمحتاج لل2,3BPG عشان يقلل ارتباط الهيموجلوبين بتاع الجنين بالاكسجين وبالتالى مستوى ال2,3BPG فدم الجنين مشعالى لانه مشمحتاجه. 

و الدم المتخزن عشان نقل الدم بيقل فيه ال2,3-BPG ولو اتنقل الدم من غيره هيعمل اختناق عشان الاكسجين مشهيسيب الهيموجلوبين وبالتالى قبل النقل بنحط فالدم inosine or glycolytic dihydroxyacetone phosphate عشان ده بيتحول الى 2,3-BPG وبعد كدة ننقل الدم

step 7:

a.  Event: (Conversion of 1,3-bisphosphoglycerate to 3­phosphoglycerate)

-  Conversion of 1,3-bisphosphoglycerate to 3-­phosphoglycerate

-  This step gproduce one ATP molecule.

b.  Enzymes stimulating this step: phosphoglycerate kinase.

هنا حد يسالنى …. احنا عندنا هنا انزيم Kinase يعنى فى مجموعة فوسفات هتتنقل طيب هى فعلا فى مجموعة فوسفات اختفت بس راحت فين؟….. الkinase هى نقل مجموعة الفوسفات من ال1,3BPG الى جزئ ADP عشان يدينا ATP  اللى هو بيتخون فيه الطاقة

Note that:

Formation of ATP from transferring a Phosphate group to ADP from organic substrate is calledSubstrate level phosphorylation.

Formation of ATP from by transferring a Phosphate group to ADP from NAD or FAD in the electron transport chain is called Oxidative phosphorylation.

Step 8:

a.   Event: (Conversion of 3-phosphoglycerate to 2­-phosphoglycerate)

Shifting the phosphate group from the carbon number 3 to the carbon number 2

a.   Enzymes stimulating this step: phosphoglycerate mutase.

The importance of this step is to make the compound suitable for the active site of the enolase enzyme.

NOTE: Mutase shifting of a functional group from one position to another within the same molecule

اهمية الخطوة ان يغير شكل المركب عشان يبقى مناسب للactive site بتاع انزيم الenolase اللى فى الخطوة الجاية ولو لاحظنا ان هنا حصل نقل مجموعة فوسفات من ذرة للتانية فنفس الجزئ يعنى تقريبا ممكن نسميها طفرة يعنى mutation يعنى هنستخدم انزيم اسمه mutase

ٍStep 9:

a.  Event: (2-Phosphoglycerate is dehydrated to form phosphoenolpyruvate)

The dehydration of the alcohol produces a double bond between carbons 2 and 3 and creates a high-energy enol phosphate linkage.

b.  Enzymes stimulating this step: enolase.

Step 10:

a.  Event: (Conversion of phosphoenol pyruvate to pyruvate)

Conversion of phosphoenol pyruvate to pyruvate producing one ATP molecule.

It is irreversible step.

b.  Enzymes Stimulating this step: Pyruvate Kinase (transfer Phosphate group from phosphoenolo-pyruvate to ADP molecule to produce one ATP Molecule)

فالخطوة دى برضو اتكون عندنا kinase يعنى هينقل مجموعة فوسفات وبالفعل هو نقل مجموعة فوسفات من الphosphoenolo-pyruvate الى جزئ ADP عشان يدينا جزء واحد من الATP.

D)  Calculation of ATP molecules from glycolysis:

We know that we get from STAGE (I) 2 molecules of glyceraldehyde-3-phosphate and we know that each molecules when continue glycolysis in the STAGE(II) give 2 ATP molecule, thus the 2 molecule of glyceraldehyde-3-phosphate give 4 ATP molecules.

Also we know that the steps number 1 and 3 consume 2 molecule of ATP

Thus, the net product of ATP during glycolysis is 2 ATP

4 ATP form Stage (II) – 2 ATP from Stage (I) = 2 ATP

المرحلة الاولى  انتهت بجزئ glyceraldehyde-3-phosphate وجزئ dihydroxyacetonephosphate اللى اتحول الى Glyceraldehyde-3-phosphate يعنى كدة المرحلة الاولى انتهت بجزيئين من الG-3-P وفالمرحلة التانية شوفنا ان كل جزئ منهم بيدينا 2ATP يعنى الجزيئن هيدونا 4ATP 

واحنا استهلكنا 2ATP فى الخطوة الاولى والتالتة يبقى هنشيلهم من العدد الكلى يعنى هيبقى 4-2 يبقى كاننا حصلنا على 2ATP فى النهاية

E)    Regulation of Glycolysis:

هنا هيتكلم على الحاجات اللى بتتحكم فى الglycolysis طبيعى ان مفيش اى سيكل فالجسم بتكون شغالة بدون تحكم

Glycolysis pathway is regulated by:

A) Control of 3 enzymes that catalyze the 3 irreversible steps of glycolysis:

  1. Hexokinase (step 1),

  2. Phosphofructokinase (step 3)

  3. Pyruvate Kinase (step 10).

هنا بيتكلم عن الانزيمات اللى بتتحكم فى الglycolysis ….. لو لاحظنا ان ال3 انزيمات دول هما ال3 انزيمات اللى مسئولين عن ال3 خطوات الirreversible وبالتالى لو اى واحد فيهم مشموجود العملية ممكن ترجع للخلف فمثلا لو الhexokinase او الglucokinase مشموجود …. مشصهيحصل phosphorylation يعنى مشهيضيف عليه مجموعة فوسفات ويحبسه ذى ماقولنا وبالتالى هيخرج الجلوكوز تانى من الخلية

B) Energy regulation:

  1. High level of ATP inhibits  phosphofructokinase (PFK-1) and pyruvate kinase.

  2. High level of ADP and AMP stimulate PFK.

اى انزيم فالدنيا لما بيزيد تركيز الناتج بتاع التفاعل اللى بينشطه بيحصله هو تثبيط يعنى مبيشتغلش وعندنا هنا الATP لما يزيد يعنى لما الخلية تنتج وتخزن كمية كبيرة من الطاقة نلاقى ان الATP بيمنع الPFK والpyruvate kinase انهم يشتغلو وبالتالى بيوقف الglyolysis وكذلك الADP  بيعمل العكس يعنى لما تكون الطاقة قليلة يعنى معنديش ATP كتير هيزيد تركيز مين الADP او بمعنى اوضح لما الخلية تستهلك طاقة هيتحول  الATP الى ADP وبالتالى هيقل تركيز الATP ويزيد تركيز الADP ولما ده يحصل بيتنشط الPFK وبالتالى دى طريقة تنظم وتتحكم فى الglycolysis

C) Substrate regulation:

  1.  Glucose-6-phosphate inhibits hexokinase (and not glucokinase).

  2. Fructose 1,6 bisphosphate stimulates phosphofructokinase-1.

  3. Citrate inhibits phosphofructokinase-1.

  4. Fructose 1,6 bisphosphate stimulates pyruvate kinase.

نفس مبدأ اللى فوق ان المادة الناتجة تتحكم فى نشاط الانزيم اللى بينشط التفاعل وممكن يكون فى مواد تانية بتتحكم فيها ذى الcitrate …. اقرا ال4 جمل

D) Hormonal regulation:

  1. Insulin: Stimulates synthesis of all key enzymes of glycolysis. It is secreted after meal (in response to high blood glucose level).

  2. Glucagon: Inhibits the activity of all key enzymes of glycolysis. It is secreted in response to low blood glucose level.

هنا عايز يقولنا ان الinsulin لما يكون الجلوكوز عالى فالدم يتفرز من البنكرياس وينشظ كل الانزيمات الرئيسية وبالتحديد ال3 اللى قولنا عليهم وده بيحصل بعد الاكل اما بالنسبة للglucagon لما يكون السكر عالى فالدم فهو يطلع ويوقف كل الانزيمات الرئيسية اللى بتعمل الglycolysis عشان لو موقفتش السكر اللى فالدم هيفضل يقل يقل وطبعا ده ممكن يعمل  اغماء

Continuing Glycolysis

تكملة اكسدة الجلوكوز

In the lecture no. 2 we saw that the end of the glycolysis is pyruvate and also NADH which is obtained in step 6, but we don’t know the fate of pyruvate and NADH; will they be waste products or will they consumed in any way? ………. this what we see in the lecture no. 3

also, we will study the importance of glycolysis and the diseases resulted from the deficiency of the 3 enzymes that catalyze the 3 irreversible steps

 فى المحاضرة التانية  شفنا ان فى نهاية ال glycolysis بيتكون pyruvate وكمان NADH فى الخطوة السادسة ولكن احنا معرفناش الpyruvate والNADH بيحصلهم ايه ….. يعنى هل بيكونو مجرد نواتج للتفاعل بتترمى وتخرج من الجسم ولا انها بتستهلك برضو؟ ….. ده اللى هنعرفه فالمحاضرة التالتة

كمان هنعرف اهمية الglycolysis وكمان الامراض الناتجة عن نقص الانزيمات المهمة الى بتنشط الخطوات الirreversible

A)  Fate of Pyruvate and NADH

We know before that in the step 5, Dihydroxyacetonephosphate (DHAP) and glyceraldehyde-3-Phosphate (G-3-P) are produced and also we know that the cell doesn’t consume DHAP, so, DHAP is converted into G-3-P……….thus,the stage 2 of glycolysis starts with 2 G-3-P and thus we get 2 NADH and 2Pyruvate from the stage 2 (the end of glycolysis) >>>>>> and we know that the conversion of DHAP to G-3-P occurs to get the energy from the 6 carbons of glucose

But also pyruvate has 3 carbon and the cell doesn’t use, …….. so, what if the fate of pyruvate? …..Will the cell get rid of it as waste product and lose the energy of 3 carbons? ….. this what we will see in this section.

in case of NADH, What is its fate? ….. it should be converted to NAD+ which is needed in the step 6 and if this conversion doesn’t occur, glycolysis can’t be repeated because there will not be NAD+……. so NADH should be oxidized through Krebs cycle and Electron transport chain or by fermentation in  case of oxygen shortage (we will study this in the next lectures).

عرفنا ان فى الخطوة الخامسة بيتكون dihydroxyacetonephosphate(DHAP وglyceraldehyde-3-phosphate وعرفنا ان الخلية مشبتسفاد من DHAP وانه بيتحول الى glyceraldehyde-3-phosphate وكدة فالمرحلة التانية من الglycolusis بندخل بجزيئنGlyceraldehyde-3-Phosphate  وfالتالى بناخد من المرحلة التانية 2NADH وكمان 2pyruvate وعرفنا كمان ان تحويل الDHAP الى G-3-P ده عشان الخلية تاخد الطاقة من ال6 كربونات بتوع الجلوكوز

طيب ما الpyruvate برضو 3 كربونات ومع ذلك الخلية لسة مستفادتش منه ………………. طيب هو كدة مصيرة ايه؟ ….. الخلية هترميه ويبقى كدة خسرت 3 كربونات ولا هيحصل فيه ايه ….. ده الهنشوفو ولاحظو اننا هدفنا اكسدة الجلوكوز الى CO2 وماء بس

بالنسبة للNADH مصيره هيكون ايه؟؟ ….. لازم يتحول لNAD+ اللى بنحتاجه فى الخطوة السادسة عشان لو ده محصلش يبقى الglycolysis هتحصل مرة واحدة بس  عشان مشهتلاقى NAD+ فلازم ناكسدها وقولنا ناكسدها ليه لانها هى طبعا حصلها اختزال فالخطوة السادسة والاكسدة دى اما تحصل فوجود الاكسجين فالkerbs cycle والelectron transport chain او تحصل فغياب الاكسجين بالfermentation ذى مهنشوف فى المحاضرات الجاية ان شاء الله.

1) Fate of Pyruvate:

Pyruvate will be oxidized to get the energy of the rest 3 carbons …. in 2 ways:

  1. Oxidation in presence of Oxygen through krebs cycle (Tricarboxylic acid cycle [TCA]) and Electron Transport chain (ETC) in Mitochondria.

  2. Oxidation in absence of Oxygen to be converted into lactate

احنا قولنا ان الpyruvate قدامه طرقين اما انه يدخل الkerbs cycle والelectrontranspot chain فى الميتوكندريا عشان يكمل اكسدة الجلوكوز فحالة وجود الاكسجين وبالتالى هيدينا كمية كبيرة من الطاقة ذى ماقولنا قبل كدة واما يتحول الىlactate فحالة غياب الاكسجين ودينا طاقة قليله

a)  Under aerobic conditions (in presence of oxygen):

In aerobic conditions, pyruvate proceeds through the tricarboxylic acid (TCA) cycle (=Kerbs cycle) to complete glucose oxidation by its conversion to acetyl CoA and oxaloacetate which are the principle intermediates for of kreb’s cycle according to the following:

i.  Oxidation to acetyl-CoA:

With presence of oxygen, pyruvate is oxidized to acetyl-CoA (acetyl coenzyme A) producing much more energy by Pyruvate dehydrogenase via oxidative decarboxylation reaction as follow:

فى وجود الاكسجين الpyruvate يبتأكسد الى AcetylCOA بواسطة انزيم pyruvate dehydrogenase وينتج كمية كبيرة من الطاقة والتفاعل ده بنسميه oxidative decarboxylation  واللى هو تفاعل اكسدة بيشيل فيه COO.

نلاحظ من المعادلة انه عمل decarboxylation يعنى شال مجموعة الCOO وحط مكانها الCoA عشان يدينا الAcetyl-CoA وسموه كده ليه عشان ذى ماحنا شايفين من تركيبه يتركب من مجموعة (acetyl) والCOA

Acetyl-CoA is a major fuel for krebs cycle as well as a building block for fatty acid synthesis in mitochondria.

الacetyl-COA ده هو المركب الاساسى اللى بيبدأ kerbs cycle وهو وحدة بناء الاحماض الدهنية

ii.  Carboxylation to oxaloacetate:

Conversion of pyruvate to oxaloacetate (OAA) by pyruvate carboxylase is an important, because it provide the citric acid cycle with OAA intermediate and provides substrate for gluconeogenesis and krebs cycle.

بيقول ان الOAA ده مهم فانه بيمد kerbs cycle بالsubstrate بتاعها عشان ذى ماقولنا نكمل اكسدة الجلوكوز الى CO2 وماء وكمان عشان الgluconeogensis ودى عبارة عن تكوين الجلوكوز من مواد مش سكرية ودى طبعا عكس الglycolysis  وهنتكلم عليها فى المحاضرة الجاية

كل اللى حصل فالتفاعل انه ضاف مجموعة COO على الCH3

b)   Under anaerobic conditions:

If no oxygen is available or in case of short supply of oxygen, cells can obtain energy through the process of anaerobic respiration such as fermentation producing lesser energy than aerobic respiration in the form of fewer ATP molecules and it is not efficient process.

فى حالة نقص الاكسجين او غيابه الخلية بتحصل على الطاقة من التنفس اللاهوائى ذى مثلا عن طريق التخمر وده بيدى طاقة اقل من اللى بناخدها فحالة التنفس الهوائى عشان فحالة اللاهوائى بنحصل على كمية قليلة من الATP وبالتالى دى عملية غير فعالة للحصول على الطاقة.

In an aerobic condition, glycolysis is repeated when the NAD+ is regenerated in the kerbs cycle and electron transport chain by oxidising NADH resulting in high energy, but in case of anaerobicrespiration glycolysis is self-functioning through regeneration of NAD+ by the conversion of Pyruvate to lactate by lactate dehydrogenase through lactic acid fermentation process without producing any ATP molecules.

احنا سبق وقولنا ان لو الNADH متحولتش الى الNAD+ يبقى الglcolysis هتحصل مرة واحدة عشان الخطوة السادسة فالglycolysis بتحتاج للNAD+ وبعد كدة الخلية متلاقيش حاجة تاخد منها طاقة وقولنا ان فى وجود الاكسجين الNADH بتتاكسد فى الkerbs cycle والelectron transport chain طيب وفى غياب الاكسجين هتاكسدها اذاى؟؟؟……….

هناكسده عن طريق تحويل الPyruvate الىlactate بواسطة انزيم lactate dehydrogenase عن طريق تخمر الlactic acid وبالتالى الglycolysis هتشغل نفسها حتى فى غياب الاكسجين يعنى كانها بتصنع البضاعة بتاعتها حتى لو التجار مرضيوش يدوها بضاعة.

This conversion of Pyruvate to lactate in anaerobic conditions is promoted by the accumulation of NADH and the depletion of NAD+ which is needed as an electron acceptor so that glycolysis can continue

يعنى عملية الfermentation اللى بنحول فيها الpyruvate الى lactate تحفز بواسطة مستوى الاكسجين فالدم وكمان  تحفز بزيادة الNADH ونقص الNAD+ وكمان العملية دى reversible وهنقول ايه فايدتها انها تكون reversible كمان شوية

In microorganisms, NAD+ is regenerated through conversion of Pyruvate to ethanol by ethanoldehydrogenase through the alcohol fermentation.

فى الكائنات الدقيقة نحصل على NAD+ خلال تحويل الpyruvate الىethanol بواسطة انزيم ethanol dehydrogenase خلال عملية تخمر الكحول (Alcohol fermentaion).

Tissues that use lactic fermentation:

1) RBCs: because it doesn’t contain mitochondria because aerobic oxidation of pyruvate occurs through Kerbs cycle and electron transport chain which occurs in mitochondria 

احنا قولنا ان الkerbs cycle والelectron transport chain بتحصل فالميتوكوندريا والNADH بتدخل فيها فى وجود الاكسجن عشان تدينا طاقة عاليا بالتالى لو الRBCs مفهاش ميتوكوندريا يبقى مشهتعمل aerobi respiration وهتعتمد فقط على الطاقة القليلة بتاعة الglycolysis فى حالة الanerobic respiration

2) Muscles: during heavy exercise the cells deplete oxygen and thus there is a shortage of oxygen supply, thus the muscle get energy through anaerobic glycolysis and regenerate NADH through the lactic acid fermentation (anaerobic respiration).

طبعا فى المجهود العالى العضلة بتحتاج طاقة عالية جدا والglycolysis هتشتغل فى وجود الاكسجين فى الاول وبسرعة كبيرة وبعد كدة الاكسجين ده هيخلص عشان سرعة الglycolysis هتخلصه والجسم مشهيلحق يمد الخلايا بالاكسجين الكافى ويبقى بالتالى هيكون فيه نقص فى الاكسجين وبالتالى العضلة هتعمل regeneration للNAD+ بالLactic acid fermentaion يعنى فى غياب الاكسجين عشان ذى ماقولنا لازم الNAD+يبقى موجود عشان الglyclysis تتكرر تانى يعنى ذى مقولنا هتشغل نفسها فعدم وجود الاكسجين وذى ماشبهناها بانها هتجيب البضاعة بتاعتها حتى لو التجار مرضيوش يدوها وبرضو قدام هنتكلم بالتفصيل عن الحكاية دى

2) Fate of NADH:

طبعا احنا لسة متكلمين عن الNADH وبتروح فين وقولنا انها بتروح فالkerbs cycle والelectron transport chain فى وجود الاكسجن اوتروح فالfermentation فغياب الاكسجين وده عشان نحصل على NAD+ عشان الglycolysis تتكرر تانى

a)  In aerobic conditions:

NADH is re-oxidized in the electron transport pathway and kerbs cycle, making 6ATP molecules in oxidative phosphorylation, thus in the aerobic respiration, the cell get 8ATP;

8ATP = 6 ATP form the electron transport chain + 2 ATP from glycolysis .

we will study kerbs cycle and electron transport chain in the next lectures.

ذى ماقولنا قبل كدة ان الNADH فى حالة وجود الاكسجين بتدخل فى الkerbs cycle والelectron transport chain فى الميتوكوندريا عشان تتاكسد وطلع NAD+ وفى نفس الوقت فالميتوكوندريا بناخد 6ATP من ال electron transport chain  وبناخد 2ATP من الGlycolysis يبقى ناتج اكسدة الجلوكوز فى وجود الاكسجين بيكون 8ATP

b)   In anaerobic conditions:

NADH is re-oxidized by lactate dehydrogenase (LDH) or alcohol dehydrogense either of which provide NAD+ for more glycolysis.

ذى ماقولنا بالظبط فوق ان الNADH بيتاكسد فى غياب الاكسجين خلال lactic acid fermentaion

B)   Interpretation of some conditions of the body:

1) Muscle cramp and muscle fatigue:

During vigorous exercise, the cell needs high energy leading to high rate of glycolysis which then consumes all oxygen in the muscle resulting in shortage of oxygen and thus it will regenerate NAD+ through the lactic acid fermentation.

But accumulation of lactate (lactic acid) in muscle increase acidity and decrease the pH causing muscle cramp شد عضلى and muscle fatigue.

So, accumulation of lactic acid is harmful for the cells, thus the cells must remove the excess lactic acid

The muscle remove the excess lactic acid when the O2 is restored by the high rapid respiration during resting stage and this occur through the lactic acid cycle (also called Cori cycle) in which the lactic acid enter the blood circulation and move to the liver in which lactate is converted to Pyruvate by the lactic dehydrogenase which then is Converted to glucose through gluconeogensis and then glucose enter the circulation to move to the muscle to give it the required energy and the cycle will repeated again in case of shortage of O2

And in the resting stage, without continuing exercise, the Pyruvate may enter the kerbs cycle and the electron transport chain in mitochondria and produce high energy

اثناء التمرين العنيف بيحصل نقص للاكسجين عشان الخلية بتحتاج طاقة عالية فال glycolysis هتكون اسرع وتستهلك الاكسجين ذى ماقولنا وبالتالى تلجا العضلة الى الlactic acid fermentation عشان تاكسد ال NADH الىNAD+ عشان تعرف تعمل glycolysis تانى وبالتالى بيتكون lactic acid كتير

لكن الlactic  acid هيتجمع وهيزود الacidity فى العضلات ويقلل الpH فيعمل شد عضلى (muscle cramp) ويخلى العضلة تتعب وبالتالى هو كدة مضر ولازم العضلة تتخلص منه بس اذاى:

هتستنى لما الاكسجين يتوفر لما الجسم يرتاح بعد التمرين وده هنلاحظه لما ترتاح بعد التمرين بتقعد تنهت يعنى تتنفس بسرعة لما توقف التمرين بس برضو ايه البيحصل لما الاكسجين يتوفر؟

الlactic acid يروح للدم ومنه للكبد وفى الكبد هيتحول الى Pyruvate اللى بعد كدة هيتحول الى جلوكوز عن طريق الgluconeogensis وبعد كدة الجلوكوز يروح للدم تانى ومنه للعضلة عشان تاخد طاقة وبعد كدة مع استكمال التمرين الاكسجين يقل والlactic acid يتكون تانى ويروح للكبد وهكذا تتكرر العملية دى ودى سيكل اسمها الCori cycle اللى هى lactic acid cycle

احنا قولنا فوق هنعرف ليه الlactic acid fermentation عمليه reversible هى كدة عشان البيتحكم فيها نسبة الاكسجين وبيتم بنفس الانزيم وهو lactate dehydrogenase

ولو بطلنا تمرين الاكسجين هيكون متوفر كتير يبقى الlactate هيتحول الى Pyruvate وده يدخل الkerbs cycle والelectron transport chain عشان يدينا طاقة وكمان لو مكانتش reversible يبقى هيحصل تجمع للlactic acid علطول وهيحصل شد عضلى علطول وطبعا ده مبيحصلش فالطبيعة

2) Wight loss of patients suffer from cancer:

The cancerous cell needs high amount of energy because it reproduce rapidly and thus glycolysis is ten times faster in solid tumors than in the non-cancerous tissue and also the solid tumors lack the blood connection and thus lack oxygen supply, thus it use the lactic acid fermentation and the Cori cycle in which the lactate formed in the tumor cell goes to the liver through the blood to converted to Pyruvate by the lactate dehydrogenase and then Pyruvate is converted to glucose through gluconeogensis to produce glucose which then pass to the blood then to the cancer cell

But here there is an energy dissipation اسراف because high energy used in the form of 6-phosphate groups to form glucose in liver to get low energy in the form of 2 phosphate groups through glucose oxidation in the cancer cells and this energy dissipation cause weight loss.

بمعنى ان الخلايا السرطانية بتنقسم بسرعة جدا وبالتالى هتحتاج طاقة عالية جدا ولقيو ان الglycolysis فالخلايا السطانية اسرع عشر مرات فالخلايا العادية وكمان الورم مشبيوصله دم وبالتالى مفيش اكسجين >>>>>> صحيح بعد شوية هيعمل لنفسه شعيرات دموية عشان يحصل على دم بس مشهيكون بالدرجة الكافية وهيكون برضو الاكسجين قليل وبالتالى هتستخدم الlactic acid fermentation وده معناه ان الlactate هيروح للكبد ويعمل الcori cycle يعنى هيروح للكبد ويتحول لpyruvate وده يتحول جلوكوز والجلوكوز يروح للدم عشان يروح تانى للخلايا السرطانية وهكذا

بس لو شفنا فالصورة بتاعة الcori cycle ان الكبد بتستخدم 6 مجموعات فوسفات يعنى 6ATP عشان تكون جزء جلوكوز واحد وفى نفس الوقت الخلية السرطانية بتستفاد بمجموعتين فوسفات بس يعنى 2ATP بس يعنى عشان نطلع طاقة من مجموعتين فوسفات بتستهلك 6 يبقى كدة احنا نسرف طاقة يعنى بنستهلك طاقة كبيرة فالتصنيع وبننتج طاقة قليلة او بمعنى تانى بنفقد calories كتير قوى وده مع الوقت بيعمل فقد فالوزن

C)   Biochemical Importance of Glycolysis

1) It is important energy source (8 ATPs) under aerobic condition or (2ATPs) under anaerobic condition. Mainly in contracting muscles, brain tissues and RBGs.

واحنا عرفنا ليه 8ATP فالaerobic respiration عشان كدة انا اجلت الbiological importance لحد مانقولها

2) It is the principle route for glucose oxidation and initiation of kreb’s cycle via pyrurate and acetyl CoA.

ودى برضو اتكلمنا عنها

3) It is the principle route for fructose and galactose metabolism.

دى لسة هنتكلم عنها لما نشوف الميتابوليزم بتاع الfructose والgalactose بس الممكن نعرفه دلوقت انهم بيتحولو لintermediate من الintermediates البتتكون فى الglycolysis وبالتالى الخلية ممكن تحصل على طاقة من الfructose والgalactose>

4) It connects between the carbohydrates, lipid and protein metabolism.

يعنى بتربط بين الميتابوليزم بتاعة الليبد والبروتين والكاربوهيدرات وهنشوف الكلام ده بعدين فى المحاضرات الجاية

5) Its reversibility is in the process of gluconeogenesis.

يعنى عشان خطواتها معظمها reversible فبتسهل تكوين الجلوكوز فالgluconeogensis فحالة الhypoglycaemia ودى هنعرفها المحاضرة الجاية

6) Glycolysis is the only energy source for RBCs and ending by lactate formation due to the absence of mitochondria

Glycolysis in Lab:

Not that, during collecting blood sample for determination of glucose level in the blood, glycolysis is still working in the RBCs in the tube and thus consuming glucose leading to false result.

Therefore, sodium fluoride is added in the blood sample to prevent enolase enzyme and inhibit glycolysis by the RBCs.

دى طبعا معظم الناس اللى بتشتغل فالمعامل عارفينها …اللى هى لما بنسحب عينة دم عشان نعمل تحليل جلوكوز فالدم .. بتكون كرات الدم الحمرا لسة شغالة وبتستهلك السكر اللى فالبلازما وبالتالى بيحطو فالانبوبة sodium fluoride عشان دى بيمنع استهلاك كرات الدم الحمرا للجلوكوز والتالى النتيجة تطلع مظبوطة ويفضل نفصل البلازما فانبوبة تانية بمجرد سحب العينة عشان ده هيقلل فرصة استهلاك الجلوكوز وفى ناس بتسحبها عادى فانبوبة مفهاش sodium fluoride بس بيفصلو السيرم بماشرة

D)  Clinical aspects of glycolysis:

1) Pyruvate kinase (PK) deficiency:-

This leads to decreasing production of the ATP which is require for the Na/K pump leading to shutting down these pumps causing malformation of the RBCs and then destruction of the RBCs and thus leading to haemolysis causing haemolytic anaemia

Excessive RBC destruction in spleen and haemolysis lead to jaundice (elevated bilirubin) and increased reticulocyte count

الRBC محتاجة الى دخول الصوديوم والبوتاسيم فيها عشان الNa/K pump القولنا عليها قبل كدة بس دى بتحتاج طاقة مفصورة  فقط على الATP وبالتالى نقص الانزيم ده بيوقف الglycolysis وبالتالى  الATP مشهيتكون والRBC مشهتلاقى طاقة والNa والK مشهيدخلو فيها فهيحصلها تشوه وتتكسر وتعمل haemolysis وتعمل haemolytic anaemia وعملية تكسير كرات الدم الحمرا بصورة كبيرة فالطحال والhemolysis بيعمل صفرا يعنى بيعلى الBilirubin فالدم وكذلك بيزيد عدد كرات الدم الحمرا الغير مكتملة التكوين اللى اسمها reticulocyte عشان طبعا النخاع لما يلاقى كمية كبيرة بتتكسر هيحاول يطلع هو كميات كبيرة وهيطلع كمية كبية منها غير مكتملة التكوين عشان يعوض النقص على قد مايقدر

2) Hexokinase deficiency:

Leads to haemolytic anaemia due to decrease ATP production. The mechanism is similar to that of PK deficiency.

دى بتعمل نفس الكلام بتاع الحالة الاولى عشان بتقلل تكوين الATP

3) Lactic acidosis:

Lactic acidosis occurs whenever production of lactate exceeds its utilization.

Lactic acidosis is characterized by a pH < 7.36 and lactate level > 5mmol/L

Lactate is produced in all tissues. Skeletal muscle, brain, red blood cells, and renal medulla are responsible for the majority of the production.

Causes of lactic acidosis:

-          It results from increased formation or decreased utilization of lactate.

-          Increased formation of lactate as in:

  • Severe muscular exercises.

  • Decreased utilization of lactate in tissues: it occurs in cases of anoxia or lack of oxygen. This is because oxygen is essential for conversion of lactate into pyruvate, which proceeds into acetyl CoA, and Krebs’cycle. Tissue anoxia may occur in conditions that impair blood flow e.g. myocardial infarction, angina pectoris, respiratory disorders,’ and anaemia.

طيعا احنا حكينا حكاية العضلات وكدة فمشهنقولها تانى ولكن هنوضح حاجة

الفرق بين الHypoxia والanoxia

الhypoxia نقص الاكسجين بس مشغيابه ولكن الAnoxia نقص الاكسجين كله

هنوضح الامراض المكتوبة

ال myocardial infarctionده عبارة عن تدمير عضلات القلب مما يؤدى الى نقص الامداد بالدم وبالتالى بالاكسجين

ال angina pectorisده الم حاد فالصدر نتيجة لعدم وصول الدم وباتالى الاكسجين للقلب نتيجة لانسداد الشريان التاجى وده بيحصل فحالة الatherosclerosis ال عرفناها باسم تصلب الشرايين

4) Oxygen Deprivation in Heart Attack and Stroke:

-          Myocardial infarction: interruption of the blood (O2) supply to a portion of the heart leading to blood clotting within the heart.

من امداد الاكسجين فجزء من القلب يؤدى الى جلطة فالقلب وتدمير خلايا القلب

-          Stroke:   interruption of the blood (O2) supply to a portion of the brain.

The main problem lack of (O2), is to block of aerobic metabolism (ETS) and induction of anaerobic metabolism (Glycolysis)  Leading to lactic acid acidosis.

المشكلة الرئيسية فى نقص الاكسجين هى توقف الميتابوليزم فى وجود الاكسجين وطبعا اللى هيحصل الميتابوليزم فى حالة نقص الاكسجين وبالتالى هيحصل تكوين للlactic acid وبالتالى يحصل ACIDOSIS

5) Consequences of O2 Limitation:

Disruption of osmotic balance (ion pumps) leading to entry of water to the cells causing Swelling of cells and organelles and increased permeability.

Acidification (anaerobic lactic acid production), high level of activity for leaked lysosomal enzymes.

 نقص الاكسجين بيعمل توقف لمخضات الايونات فى جدار الخلايا وده هيسمح بدخول كميات كبيرة من الماء ويخلى الخلية تتنفخ وكذلك العضيات الصغيرة اللى جوة الخلايا ويزود نفاذية اغشية الخلايا

كمان بيزود الACIDOSIS ودى قولناها الف مرة

 How to diagnose Myocardial Infarction in lab:

-       The lactate dehydrogenase had 5 forms, LDH 1,2,3,4, and 5

-     The LDH1 is the present in the heart and it presence is indication to lactic acid formation and so, indication of shortage of (O2) supply.

LDH5 is induction to hepatitis.

 بنعرف جلطة القلب فى المعمل عن طريق كمية الlactate dehydrogenase فالدم …. وده ليه 5 انواع …LDH1,2,3,4,5 

 LDH1 موجود فالقلب … ولو لقيوه يبقى معناه ان الlactic acid بيتكون وبالتالى ده اشارة الى نقص امداد الاكسجين.

 LDH5 وجوده فالدم معناه ان المريض عنده hepatitis

بناءا على طلب الناس ال اشتكت من حفظ اسامى الانزيمات انا هبسطهالكو

الkinase معناها انه بينقل مجموعة فوسفات من جزئ للتانى سواء من المركب لل ATP او العكس وده بيحصل فى خطوات الphosphorylation وهنلاقى اسمه من اسم الsubstrate ذى الglucokinase معناه انه بينقل مجموعة فوسفات من الATP للglucose والphosphofructokinase معناه انه بينقل مجموعة فوسفات من الATP لفركتوز موجود فيه اصلا مجموعة فوسفات

الmutase ده بينقل مجموعة فوسفات من مكان للتانى ذى الphosphoglycerate mutase معناه انه بينقل مجموعة الوفسفات اللى فىphosphoglycerate على كربونة تانى فى نفس الجزئ ذى فالخطوة 8

الisomerase يعنى بيحول ايزومير للتانى ذى تحويل الDHAP لGAP فى الخطوة رقم 5 ومحدش يقولى ما هنا نقل الفوسفات فى نفس المركب  ….. بس هو حول الالدهيد لكيتون يعنى مشنفس المركب لكن عمل isomerisation

الadolase ال فى الخطوة 4 ده اسمه جاى من التفاعل العكسى بتاع تحويل الDAHP and GAP الى F-1,6-bisphosphate عشان تفاعل كيتون مع الدهيد اسمه aldol condensation فسموه aldolase

الenolase عشان التفاعل ده بيعمل enol bond اللى هى كدة (CH2=C-O) ودى موجودة فالphosphoenolopyruvate
